These modifications play critical roles in regulating chromatin structure and gene expression.

GPCRs: Exploring New Paradigms

As one of the largest and most diverse protein families in nature, the G-protein coupled receptor (GPCR) superfamily play important roles in a variety of biological and pathological processes such as development and proliferation, neuromodulation, angiogenesis, metabolic disorders, inflammation, and viral infection. Not surprisingly, it is one of the most targeted protein families in pharmaceutical research today (Schlyer 2006).
All members of this superfamily share a similar seven transmembrane domain, but are grouped on the basis of shared sequence motifs into the following broad classes:
Class A Rhodopsin-like
Class B Secretin-like
Class C Metabotropic glutamate/pheromone
Class D Fungal pheromone
Class E cAMP receptors
Frizzled/Smoothened
Vomeronasal receptors
Putative/unclassified
The pharmaceutical industry has focused on development of modulators to this protein family, but GPCRs also represent an attractive target as biomarkers. Recent studies have implicated endothelin receptors, chemokine receptors and lysophosphatidic acid receptors in tumorgenesis and metastasis, and a recent in silico analysis found a number of GPCRs that were up-regulated in various cancers which suggests not only targets of therapeutic value, but of diagnostic and prognostic value as well. US prepares to act

The US, the largest greenhouse gas emitter in the world, may soon be picking up the challenge of address climate change.
Senator John Kerry, en route to international climate change talks in Poland said, “It’s a very exciting time. It’s a moment we have been waiting for, many of us, for some period of time; we intend to pick up the baton and really run with it.”
President-elect Obama has been “very, very clear that after eight years of obstruction and delay and denial, the US is going to rejoin the world community in tackling this global challenge,” added Senator Kerry who will be an observer at the December 1st to 12th Conference.
Kerry said the US message will be “that America is back, we are back in a position of participation, of respecting views and having real discussions and trying to find the best framework for all of us.”
The United States is currently the only industrialized nation not to ratify the emission-capping Kyoto Protocol.
Obama vowed he would “engage vigorously” in global climate change talks in a video message to an international conference in California.
“Now is the time to confront this challenge once and for all. Delay is no longer an option. Denial is no longer an acceptable response. The stakes are too high. The consequences, too serious,” said Obama.
“For the first time in many years you will have an administration prepared to negotiate a commitment on the part of the US. Without that there are no prospects for an agreement,” said Elliott Diringer, vice president for international strategy at the Pew Center for Global climate change. “Will the world will be disappointed, that depends on expectations and I think expectations in some places are too high. But I think the president elected sent a very clear signal of what to expect in his statement last week when he said returning US emissions to 1990 levels by 2020. That is what he will try to deliver. It won’t be easy but it can be done.”
Two climate change bills will go before the US Senate in January.
Barbara Boxer, chairwoman of the Senate Environment and Public Works committee, said one bill would combat harmful gas emissions by providing 15 billion dollars a year to spur clean energy innovation and the development of advanced biofuels.
The other bill seeks to direct the US Environmental Protection Agency to establish a cap-and-trade system to limit greenhouse gas emissions.
